Gel filtration (also known as size exclusion chromatography (SEC)) be divided into two main areas:
1) High-resolution purification of biomolecules
2) Group separations
To get working knowledge of gel filtration for purification of proteins please read and digest the following chapters in the handbook 'Size Exclusion Chromatography'. Introduction (page 7-8), chapter 1, chapter 2, chapter 5, and chapter 7.
