Chemical Biology and Medicinal Chemistry

The research in Mads Hartvig Clausen’s group focuses on chemical biology, using synthetic chemistry to develop tool compounds that address key biological questions. This overall research theme manifests in several focus areas, including synthetic methodology, medicinal chemistry, and molecular design. The aim here is to create small-molecule libraries, prodrugs, biologically active compounds, drug delivery strategies, vaccine development, and studies of complex biomolecules such as plant cell wall polysaccharides. We are part of the Chemical Biology unit of Organic Chemistry.

Research

Our research activities include the synthesis of oligosaccharides related to plant cell wall glycans, which are used to characterize monoclonal antibodies and enzymes. We also design and synthesize shape-diverse, sp3-rich high-throughput screening (HTS) compounds and fragments, enabling exploration of chemical space for drug discovery.

 

A further focus is the development of prodrugs activated by reactive oxygen species (ROS) for targeted treatment of inflammatory diseases. Furthermore, in combining synthetic design with biological evaluation, we contribute to carbohydrate-based vaccine development.

We are grateful to the DTU POC fund for their support of our vaccine technology. The funding will enable us to acquire instrumental in vivo data and strengthen our business plan for Vaccinologics.

Mads will be at the ACS Spring conference in Atlanta. He will be presenting recent development in our carbohydrate-based anticancer vaccines in the CARB division and catching up with collaborators.

Special welcome to Dr. Signe Skadborg – we look forward to the collaboration with you and the group of Professor Sine Reker Hadrup.

Congratulations to Cláudia and Faranak. Our collaboration with the Lehtiö lab in Oulo University is out in Structure. The paper describes the development of selective PPI inhibitors for the ARC4 domain, one of five ARCs in tankyrases. This has been an exemplary multi-site program enabled by EU-OPENSCREEN DRIVE – and we look forward to continuing our collaboration and exploring this compound class in the future.

Welcome to Andreas Kissling Harris. He teamed up with us through the Open Entrepreneurship co-founder program and will be instrumental in maturing our vaccine technology towards incorporation.

We kicked off the DTU Immunology Forum on January 15. Exciting day of great science – it was inspiring to witness the breath of immunology research going on at DTU.

We are grateful to the Novo Nordisk Foundation for supporting the DTU Screening Core through their upgrade program! Thanks to NNF support, DTU SCore 2.0 will include high-content screening – we look forward to implementing this for our users and collaborators during 2026.

One position will be focused on complex oligosaccharide synthesis of structures from the plant cell wall glycan RG-II. The group was well represented at the 2nd symposium by the Society for Medicinal Chemistry and Chemical Biology. Several posters were presented, and Dileep had a flash presentation.

 

 

The annual meeting in the Danish Society for Carbohydrate Chemistry was held at the University of Copenhagen. Great talks by Danish carbohydrate chemist and our invited speaker, Prof. Matthieu Sollogoub. At the event, Mads was presented with the prize for “Carbohydrate Chemist of the Year, 2024” – he is very honored and grateful to the society for the award.


 


Mads was in Clearwater Beach, FL, presenting our work on fluorinated fragment libraries for screening protein and RNA targets. The 3F and ROFF research was well received at the ACS Winter Fluorine Conference.
